Advances in developing tris(8-quinolinolato)gallium(III) as an anticancer drug: critical appraisal and prospects
Andrei R. Timerbaev,  Metallomics, 2009, 1, 193
DOI: 10.1039/b902861g

This minireview highlights advances in the development of one of the most promising gallium-based drugs to fight cancer and a critical point of view on the rational pathway to follow in the discovery of more effective anticancer weaponry.

Perspective

Zinc and tetrathiomolybdate for the treatment of Wilsons disease and the potential efficacy of anticopper therapy in a wide variety of diseases
George J. Brewer,  Metallomics, 2009, 1, 199

Two new anticopper drugs developed by the author, zinc and tetrathiomolybdate, effective in Wilsons disease, are discussed. Lowering copper levels with tetrathiomolybdate has antifibrotic, antiinflammatory and anti-autoimmune efficacy.

Critical Review

Interplay of metal ions and urease

Cells use an intricate assembly system to create the dinuclear nickel active site of urease that catalyzes the deceptively simple decomposition of urea.

This tutorial review discusses the current evidence for the possible epigenetic mechanisms for nickel, arsenite, chromium and cadmium and how these changes may play a role in their metal-induced carcinogenicity.

Consideration of the in vivo and in vitro metal-binding trends of ca. 20 recombinant metallothioneins from divergent organisms coincides in a new classification: a stepwise gradation between Zn- and Cu-thioneins where the intermediate positions in the list are polyvalent MTs of merging Zn-/Cu-character.

Differential binding of apo- and holo-FbpA with TbpA was established for the first time by SUPREX. Iron stripping from holo-Tf by either TbpA or TbpB was observed and a detailed model for iron transport is proposed.
