Bicyclic 2-pyridones act as anti-adhesive agents, blocking the complex molecular machinery of pilus biogenesis by binding the periplasmic chaperone PapD. They have been designed to target the invariant active site residues R8 and K112 in chaperones, which are key-proteins in pilus assembly. The work was carried out by Fredrik Almqvist and co-workers at Umeå University, Sweden.
