Cumulus is removed from a bovine zygote using the microfluidic technique used in these experiments. First, the bulk cumulus is removed with a constant negative pressure in the removal port (bottom vertical channel). Pressure fluctuations are then used to remove the remaining cumulus with minimal force. The top removal port (horizontal channel, right middle) draws detached cumulus out of the main channel. The main channel is 500 μm x 500 μm (W x H). The removal ports are 30 μm x 70 μm (W x H). Fluid is manually controlled with a 1 cc syringe subsequent to each removal port.
